If you click on "Edit" next to an Associated Product in a Configurable Product's edit window, when you save that simple product, it will no longer show up as associated to that parent configurable product (ie, breaks the association).

In addition, if you edit the simple product manually (ie, not through the pop-up but through a full edit) and change "Manage Stock" to be No, the Configurable Product will still show "Not In Stock" when looking at that product through Associated Products tab.
