Will upgrading to the latest release of Magento wipe out any design changes I’ve made or wipe out the product listing?

(I hope by design changes you are referring to the changes made to layout, template files)
You changes will not be wiped out if you have a custom design interface ... the default design interface will be overwritten with changes on the new version.
